The One-Step Synthetic Cannabinoids Spice Drug Screen
Test is a lateral flow immunoassay for the specific, qualitative detection of
synthetic cannabinoids metabolites in human urine at a cut-off level of 30
ng/ml. The synthetic cannabinoids detected by
the test include, but are not limited to, the metabolites of JWH-018 and
JWH-073. This assay is intended for forensic use only. This assay provides only
a preliminary result. Careful consideration and professional judgment must be
applied to any drug of abuse test result, particularly in evaluating a
preliminary positive result. In order to obtain a confirmed analytical result, a
more specific alternate chemical method is needed.
Note: This assay provides only a preliminary analytical test result. A more specific alternative chemical method such as high-pressure liquid chromatography (HPLC)
or Gas chromatography/mass spectrometry (GC/MS) must be used in order to obtain
a confirmed analytical result. Clinical consideration and professional judgment should be applied to all test results, particularly when preliminary positive results are indicated.

SUMMARY AND EXPLANATION OF THE TEST
Synthetic Cannabis (Spice) is a family of compounds
that when consumed mimics the effects of Marijuana. It is also known by the
brand names of K2 and Spice, both of which have largely become trademarks used
to refer to any synthetic cannabinoids product. Studies suggest that synthetic
cannabinoid intoxication is associated with acute psychosis, and the worsening
of previously stable psychotic disorders among vulnerable individuals such as
those with a family history of mental illness. JWH-018 and JWH-073 are the
primary synthetic cannabinoid receptor agonists responsible for the euphoric and
psychoactive effects that imitate Marijuana and are among the numerous compounds
found in “herbal” incense or smoke blends. Most popular herbal smoking products
are marketed under the brand names of K-2, K-3, Spice, Genie, Black Mombo, Pot-pouri,
Buzz, Pulse, Hush, Mystery, Earthquake, Ocean Blue, Stinger, Yucatan Fire, as
well as many others.
The excretion half-life of Synthetic Cannabinoids
(Spice) metabolized in urine, estimated by
different techniques, ranges from about two to ten days
depending on a number of factors including frequency of use. The One-Step
Synthetic Cannabinoids Drug Screen employs a unique combination of monoclonal
antibodies to selectively identify JWH-018 and JWH-073 in urine with a high degree of
sensitivity at 30 ng/ml, the established cut-off level for
synthetic cannabinoids The test device is a rapid, visually read screening test that does not require instrumentation and provides a fast indication of recent
synthetic marijuana consumption.

PRINCIPLE
 The
One-Step Synthetic Cannabinoids (Spice) Drug Screen
Test is based on the principle of competitive immunochemical reaction between a
chemically labeled drug (drug-protein conjugate) and the drug or drug
metabolites which may be present in the urine sample for the limited antibody
binding sites. The test contains a nitrocellulose membrane strip pre-coated with
drug-protein conjugate in the test region and a pad containing colored
antibody-colloidal gold conjugate. During the test, the urine sample migrates
upward and rehydrates the antibody-colloidal gold conjugate. The mixture then
migrates along the membrane chromatographically by the capillary action to the
immobilized drug-protein band on the test region. When drug is absent in the
urine, the colored antibody-colloidal gold conjugate and immobilized
drug-protein bind specifically to form a visible line in the test region. When
drug is present in the urine, it will compete with drug-protein for the limited
antibody sites. The line on the test region will become less intense with
increasing drug concentration. When a sufficient concentration of drug is
present in the urine, it will fill the limited antibody binding sites. This will
prevent attachment of the colored antibody-colloidal gold conjugate to the
drug-protein on the test region. Therefore, the presence of the line on the test
region indicates a negative result for the drug and the absence of the test line
on the test region indicates a preliminary positive
result for the drug. A visible line generated by a different antigen/antibody
reaction is also present at the control region of the test strip. This line
should always appear, regardless of the presence of drugs or metabolites in the
urine sample. This means that a negative urine sample will produce both test
line and control line, and a positive urine sample will generate only control
line. The presence of control line serves as a built-in control, which
demonstrates that the test is performed properly.

ASSAY
PROCEDURE
-
Collect a urine sample from test subject using a suitable clean container preferably glass.

Refrigerated specimens or other materials should be equilibrated to room temperature before testing.

Open the foil pouch at the notch, remove the test device, and label the device with specimen ID.
Remove test cap.

Holding the test vertically, dip
the wick end into the sample urine. Do not dip past the line indicated on the
top of the test wick .

Observe migration, or lateral flow of the sample across the entire test
panel.

Positive results may be observed as soon as
2-3 minutes, depending on the concentration
of Synthetic Cannabinoids in the tested specimen.
Read results at 5 minutes. Do not interpret results after 60
minutes. If a permanent record is required, a scan or photo copy of the
completed test device can be made.

development of two lines, a control line and a test line, no matter how
faint, in the test result window indicates a negative test screen for
the targeted drug or drug metabolite.

development of only one line, the control line, in the test result
window indicates a positive test screen for the targeted drug or drug
metabolite

The
absence of a control line in the test result window indicates an invalid
test result regardless of the presence of a test line in the test result
window. Generally an invalid result is the result of
insufficient sample being absorbed.

Positive: One (1) pink-rose band appears in the control zone and no band appears
in the test zone. A positive result indicates the presence of
Synthetic Cannabinoids
(Spice) at a level of 30 ng/ml or higher in the test urine sample.
Negative: One band appears in the test zone and one band appears in the control zone. A negative result indicates
the presence of
Synthetic Cannabinoids
(Spice) at a level of zero or less than
30 ng/ml in the test urine sample.
IMPORTANT: any line, no matter how faint, appearing in the test area confirms a negative test.
Invalid: If there are no distinct color bands visible in both the test zone and the control zone or if there is a visible band in the test zone but not in the control zone, then the test is invalid. In this instance, re-testing of the specimen is recommended.
